Stem Cell Technology and Cancer Biology Laboratory
Stem cell Technology is the Holy Grail of therapeutic intervention, offering a plethora of unique applications. I use cell signalling approaches to characterize and validate this technology in the context of disease models especially cardiovascular disorders .
My early research career at the Medical College of Wisconsin demonstrated for the first time the capacity of EETs to enhance human endothelial cell survival by inhibiting both the intrinsic, as well as extrinsic, pathways of apoptosis, an important underlying mechanism that may promote angiogenesis and endothelial survival during atherosclerosis and related cardiovascular ailments. This contribution to the protection of cardiomyocytes is highly significant because of its importance in the medical field especially in the treatment of diseases. This came as a spin-off for my interest in cardiomyocyte regeneration and interest in stem cells as a potential therapeutic agent.
I had the honor and priviledge of being trained in the lab of WiCell , a unit of Dr James Thomson , in Wisconsin and that is when stem cell research enthused me and since then I have been working in Stem cell technology in collaboration with Dr John Lough, Dr Zeljko Bosjnak from Medical College of Wisconsin. I also was awarded a DST- UKIERI project in Collaboration with Dr Eustace Johnson , University of Chester, U.K . My lab at Center for Biotechnology, Anna University is mainly focused on basic research in stem cells and differentiating them towards cardiac lineage and research also focusses on Cancer Stem cells. We are successful in isolating adult cardiac stem cells (CSCs) from a rodent model. CSCs were differentiated to cardiomyocytes using combination of epigenetic modifier - 5-Azacytidine and small molecule – Ascorbic acid. Differentiated cardiomyocytes were structurally and functionally analyzed for derivation of cardiomyogenic lineage. Studies on transplantation of differentiated cardiomyocytes in small animal myocardial injury model are being planned.
Hence my overall interest in cardiovascular research emanates from the point that there is no effective strategies or therapeutics to combat this disease. Having worked in cardiomyocyte cell death and ischemic injury for more than twenty years and I am beginning to understand the mechanisms and working on therapeutics and cell repair and replacement by stem cell therapy which might protect the cardiomyocyte apoptosis and cardiac repair in cardiovascular disorders.

Cardiovascular Therapeutics
• In vitro studies in enhancing CSCs survival, proliferation and subsequent differentiation to functional Cardiomyocytes under hypoxic conditions.
• Analysis of anti-apoptotic and cyto protective pathways give clue regarding molecular mechanisms involved in decreasing the extent of cell death.
• Isolation, culture and characterization of CSCs from rat heart biopsies by explant method
Cancer Biology
• Understanding the signaling mechanisms in Cancer stem cells
• Mitochondrially targeted compounds novel therapeutic agents for Cancer novel therapeutic agents for Cancer
• Tumour Angiogenesis
Our goal is to understand the mechanism of tumor derived cytokines recruits and regulates the fate of infiltrating stem cells and how these cells affect tumor progression. We are focused in genetic and metabolic profiling that occur in tumor and MSCs.
